Le microglia regolano la crescita e l'integrità della mielina del sistema nervoso centrale

La mielina è necessaria per la funzione degli assoni neuronali nel sistema nervoso centrale, ma i meccanismi che supportano la salute della mielina non sono chiari. Sebbene i macrofagi nel sistema nervoso centrale siano stati implicati nella salute della mielina1, non è noto quali popolazioni di macrofagi siano coinvolte e quali aspetti influenzino. Qui mostriamo che le microglia residenti sono cruciali per il mantenimento della salute della mielina in età adulta sia nei topi che negli esseri umani. Dimostriamo che le microglia sono superflue per il rivestimento mielinico dello sviluppo. Tuttavia, sono necessari per la successiva regolazione della crescita della mielina e della funzione cognitiva associata, e per preservare l’integrità della mielina prevenendone la degenerazione. Mostriamo che la perdita della salute della mielina dovuta all'assenza di microglia è associata alla comparsa di uno stato di oligodendrociti mielinizzanti con metabolismo lipidico alterato. Inoltre, questo meccanismo è regolato attraverso l’interruzione dell’asse TGFβ1–TGFβR1. I nostri risultati evidenziano le microglia come promettenti bersagli terapeutici per condizioni in cui la crescita e l'integrità della mielina sono disregolate, come nell'invecchiamento e nelle malattie neurodegenerative2,3.
La mielina riveste gli assoni neuronali per garantire la loro salute e la rapida propagazione degli impulsi elettrici per supportare le funzioni del sistema nervoso centrale (SNC), ad esempio la cognizione. L'apprendimento e la memoria implicano la formazione di mielina e richiedono che la mielina abbia una buona integrità strutturale. Gli strati di mielina sono compattati ad uno spessore proporzionale al diametro dell'assone4; tuttavia, con l'invecchiamento e nelle malattie neurodegenerative, la distruzione di queste proprietà della mielina avviene attraverso l'ipermielinizzazione. Aree allargate di mielina non compattata (dove cresce la mielina) portano a una mielina più spessa, che si disfa e forma protuberanze (chiamate outfolding) e in questi contesti si verifica anche la perdita dell'integrità della mielina attraverso la degenerazione2,3,5. Questi cambiamenti nella mielina portano a disturbi cognitivi nei topi e predicono scarse prestazioni cognitive nei primati non umani anziani e negli esseri umani6,7,8,9. Tuttavia, i meccanismi fondamentali che coordinano la formazione, la crescita e l’integrità della mielina del sistema nervoso centrale non sono chiari. Ricerche recenti hanno coinvolto in questo processo una popolazione di macrofagi residenti nel sistema nervoso centrale, le microglia. La mielinizzazione e la generazione di oligodendrociti che formano la mielina sono compromesse in seguito alla deplezione della microglia attraverso la perdita della funzione del recettore del fattore 1 stimolante le colonie pro-sopravvivenza (CSF1R)1. Tuttavia, questo approccio prende di mira anche i macrofagi associati al confine residenti nel sistema nervoso centrale (compresi i macrofagi perivascolari) e i monociti del sangue. Pertanto, non è chiaro quali popolazioni di macrofagi regolino la mielina e non è noto il coinvolgimento specifico delle microglia nella formazione e nella salute della mielina.
Per rispondere a queste domande, abbiamo utilizzato un modello di topo transgenico recentemente sviluppato in cui il super-potenziatore dell'elemento regolatore intronico Fms (Fire) del gene Csf1r (FireΔ / Δ; Fig. 1a) viene eliminato. Questa delezione porta all'assenza di microglia dallo sviluppo (quando normalmente emergono) fino all'età adulta, mentre sono presenti altri macrofagi del sistema nervoso centrale10,11. Questi topi non presentano molti dei problemi confondenti che si verificano in altri modelli con deficit di microglia, come morte dello sviluppo, anomalie ossee e assenza di macrofagi e monociti perivascolari del sistema nervoso centrale10. Utilizzando il marcatore microgliale TMEM119, abbiamo confermato che le microglia erano impoverite nel più grande tratto di sostanza bianca del cervello, il corpo calloso (Fig. 1b). I pochi macrofagi IBA1 + conservati nei topi FireΔ / Δ erano positivi per il marcatore macrofagico perivascolare LYVE1 (Fig. 1c, d). Inoltre, la presenza di queste cellule è stata confermata dall'osservazione di cellule CD206+ adiacenti ai vasi sanguigni CD31+ (Dati estesi Fig. 1a). Le densità dei macrofagi perivascolari non erano significativamente alterate nei topi FireΔ / Δ in questo momento (Fig. 1e) o in età avanzata (Dati estesi Fig. 1b, c) nonostante la ridotta espressione di CSF1R (Dati estesi Fig. 1d-f). Numeri simili di astrociti (GFAP+SOX9+) sono stati osservati nel corpo calloso dei topi FireΔ/Δ e dei compagni di figliata Fire+/+ (cellule GFAP+SOX9+; Dati estesi Fig. 1g,h). In particolare, a un'età in cui la mielinizzazione è in corso (dal giorno 25 postnatale (P25) a P30), i topi FireΔ / Δ avevano generato oligodendrociti maturi (OLIG2 + CC1 +) (Fig. 1f, g), che formavano una proporzione simile del lignaggio degli oligodendrociti (OLIG2+) come in Fire+/+ littermates (Fig. 1h). La mielina si è formata nei topi FireΔ / Δ, come indicato dall'espressione delle proteine mieliniche MAG, MBP, CNPasi, MOG e PLP nel corpo calloso (Fig. 1i e Extended Data Fig. 2a-c) e nel cervelletto (Extended Data Figura 2d,e). L'analisi ultrastrutturale ha confermato che la mielinizzazione procedeva nei topi FireΔ/Δ (Fig. 1i), senza differenze significative nel numero di assoni mielinizzati rispetto ai topi Fire+/+ (Fig. 1j) indipendentemente dal diametro dell'assone (Fig. 1k). I nostri risultati indicano che le microglia sono superflue per la maturazione degli oligodendrociti e il rivestimento mielinico dello sviluppo. Questa scoperta è in contrasto con le precedenti attribuzioni di queste funzioni alla microglia in seguito all’esaurimento di tutte le popolazioni di macrofagi del sistema nervoso centrale.
